Cat.No.:D32011A
Price: ¥
Specifications:
This product has optimized the seamless cloning buffer system compared to the fixed-point mutation kit. It is a kit that can introduce point mutations, insertions or deletions into any type of plasmid DNA. The optimized system significantly improves the accuracy of multiple point mutations and can also be used for single-site mutations.
This kit provides DH5α glycerol bacteria, which can be used for the preparation of competent cells.
Low template usage: Exponential amplification
Simple operation: No need for special carriers, restriction sites or target plasmid methylation status, no need for separate steps to destroy the initial template
Widely applicable: Suitable for multiple-site mutations and single-site mutations, with amplification length up to 10kb

This product can introduce point mutations, insertions or deletions into any type of plasmid DNA, achieving multiple point mutations and single point mutations of the plasmid DNA.
How to choose the single-site mutation kit and the multi-site mutation kit?
The PCR Mix and recombinase in the multiplex mutation kit have been optimized. They offer higher mutation success rates for mutations with ≥ 4 sites, as well as for high GC fragments and long fragments (≥ 10 kbp). If the number of mutation sites is small and the fragment length is moderate, the site-directed mutation kit can be chosen.
How to increase the success rate of multiple mutations?
The success rate will be affected by the following factors: · The distance between the mutation sites (sites that are farther apart are more efficient); · The type of mutation (base substitution is more efficient than insertion/deletion); · The size of the plasmid and the complexity of the sequence; It is recommended to conduct a small-scale preliminary experiment before the formal experiment to evaluate the efficiency.
